The TRizon RNA Purification Kit provides a simple, reliable, and rapid method for isolating high-quality total RNA from a variety of samples, including animal and plant cells and tissues, bacteria, and yeast. This kit takes advantage of the powerful lysis capabilities of TRizon reagent.

TRizon reagent preserves RNA integrity while disrupting cells and lysing cellular components. Chloroform was added followed by centrifugation to separate the solution into aqueous and organic phases. RNA remains only in the aqueous phase.

After transferring the aqueous phase, RNA was recovered by precipitation with isopropanol. After removal of the aqueous phase, DNA and proteins in the sample can be recovered by continuous precipitation. Ethanol precipitation yields DNA from the intermediate phase and isopropanol precipitation yields protein from the organic phase.

Co-purification of DNA may help to normalize RNA yield between samples.

Total RNA isolated with TRizon reagent is free of protein and DNA contamination. It can be used in Northern blot analysis, dot blot hybridization, poly(A)+ selection, in vitro translation, RNase protection assays, and molecular cloning.

Precautions to Prevent RNase Contamination RNases can be accidentally introduced into RNA preparations through improper technique at any point during the isolation process. Since RNase activity is difficult to inhibit, its introduction must be prevented. The following guidelines should be followed when working with RNA. Always wear disposable gloves. Skin often harbors bacteria and molds that can contaminate RNA preparations and be a source of RNases. Practice good microbiological techniques to prevent microbial contamination. Use sterile single-use plastic utensils and automatic pipettes reserved for RNA work to prevent cross-contamination with RNase in shared equipment. For example, a lab using RNA probes may use RNase A or T1 to reduce background on filters, and any non-disposable items such as automatic pipettes may be a rich source of RNase
