Suspend 22.0 g of medium in one liter of distilled or deionized water. Mix well and heat stirring frequently until medium dissolves. Sterilize in autoclave at 121°C (15 lbs) for 15 minutes.

Principle and explanation:

Mueller Hinton broth is low in thymine and thymidine. Only calcium and magnesium ion concentrations can be used for broth dilution antimicrobial susceptibility testing. Mueller Hinton Broth Thymine and Thymidine Content Mueller Hinton Broth is not formulated to have low thymine and thymidine content. Can be used for general bacterial culture

Acid hydrolysates of casein and beef provide nitrogenous compounds, carbon, sulfur and other essential nutrients. Starch acts as a protective colloid against toxic substances present in the medium. Starch is hydrolyzed to produce glucose as a source of energy.

Dehydrated medium is a free-flowing pale yellow powder.

The prepared medium is a light yellow transparent liquid.

Precautions:

This media is for laboratory use only.

Dry media that has exceeded its shelf life, has clumps, or has changed color cannot be used.

Storage conditions and shelf life:

Mueller Hinton agar must be stored sealed in the original container at 5-30°C.

Dehydrated medium has a shelf life of 3 years from the date of production.

The prepared culture medium can be stored in the dark at 2-8°C.

QC:

Observe culture characteristics after culturing at 35 -37°C for 18 -24 hours.
